D-Luciferin Potassium Salt

Luciferin is a chemical substance found in the cells of various bioluminescent organisms. When luciferin is oxidized under the catalytic effects of luciferase and ATP, a bluish-green light is produced. Because the reaction is dependent on ATP, it allows researchers to determine the presence of energy or life. Firefly luciferin is a particularly good reporter for in vivo biophotonic imaging due to properties of its emission spectra.

D-Luciferin Potassium Salt (P/N 122796) is isolated from firefly and extensively validated in a number of biophotonic imaging applications using the IVIS® Imaging System available from Caliper Life Sciences.

HT RNA LabChip® Assay Kit

The HT RNA assay kits provides a fast and easy way to analyze RNA samples ranging in size from 100 to 6,000 nucleotides. Performing RNA sample analysis with the HT RNA assay kit saves time and money by automating steps for quantitation and integrity analysis. HT RNA provides the user with RNA concentration and ribosomal ratios as an indicator of integrity.

The LabChip 90 Electrophoresis System uses a single sipper microfluidic chip to aspirate RNA samples directly from 96- or 384-well plates. The microfluidics chip technology automatically mixes in an intercolating dye*, electrophoretically separates, and analyzes the RNA sample. The instrument optics detect the laser-induced fluorescent signal. System software automatically analyzes the data and determines fragment size and concentration using ladder and marker calibration standards. Digital data results are immediately available for review or reporting in virtual gel, electropherogram graph, or table summary form.

The IVIS Spectrum is the newest imaging system available from Caliper Life Sciences. Like previous IVIS systems, it uses Xenogen’s novel patented optical imaging technology to facilitate non-invasive longitudinal monitoring of disease progression, cell trafficking and gene expression patterns in living animals.

The IVIS Spectrum is the most versatile and advanced in vivo imaging system available on the market today. An optimized set of high efficiency filters and spectral un-mixing algorithms lets you take full advantage of bioluminescent and fluorescent reporters across the blue to near infrared wavelength region. It also offers single-view 3D tomography for both fluorescent and bioluminescent reporters that can be analyzed in an anatomical context using our Digital Mouse Atlas.

For advanced fluorescence imaging, the IVIS Spectrum has the capability to use either trans-illumination (from the bottom) or epi-illumination (from the top) to illuminate in vivo fluorescent sources. 3D diffuse fluorescence tomography can be performed to determine source localization and concentration using the combination of structured light and trans illumination fluorescent images. The instrument is equipped with 10 narrow band excitation filters (30nm bandwidth) and 18 narrow band emission filters (20nm bandwidth) that assist in significantly reducing autofluorescence by the spectral scanning of filters and the use of spectral unmixing algorithms. In addition, the spectral unmixing tools allow the researcher to separate signals from multiple fluorescent reporters within the same animal.
LabChip® EZ Reader II

The LabChip EZ Reader is a breakthrough benchtop instrument which facilitates kinase testing and other enzyme screening and profiling. When combined with ProfilerPro kits, a low cost, turn-key solution of lab-bench kinase panel profiling is provided. LabChip assays are separations-based, so the quality of results exceeds what is achievable in homogenous, well-based assays. The LabChip EZ Reader runs kinase and enzymatic assays with a 4-Sipper microfluidics chip.

Utilizing Caliper’s widely adopted mobility shift microfluidics assay technology, the LabChip EZ Reader analyzes kinase activity by ‘sipping’ stopped reactions from 96 or 384 well microtitre plates. Caliper’s Mobility Shift Assay technology is designed for in-vitro enzyme assays aimed at the drug discovery process. It combines the advantages of capillary electrophoresis and microfluidics providing direct measurement of substrate and product, rapid assay development and real time kinetic reads.

The EZ Reader is the perfect platform for running kinase profiling with the Caliper Profiler Pro Kits. Profiler Pro Kinase Profiling kits consist of matched pairs of 384 well kinase assay and substrate plates. The assay plates each contain 24 different kinases, pre-dispensed and frozen. The substrate plates contain the appropriate fluorescent labeled peptides and ATP.

The LabChip EZ Reader II provides the option to run assays with either a 4-Sipper or a 12-Sipper microfluidics chip, providing a complete solution for developing and running enzymatic assays.

Using LabChip EZ Reader II, researchers can monitor enzyme reactions over time, resulting in more detailed data resulting in using less reagents. The advantages of microfluidics technology with real time kinetic analysis include: simplified and accelerated assay development and optimization, calculation of Km and Ki values from initial rates, and determination of enzyme linearity.
This plasmid is a shuttle vector that will replicate in a range of Gram-positive and Gram-negative bacteria. However, amplification of this plasmid in E. coli will induce mutations and make the plasmid unstable. Thus, we recommend using the plasmid supplied from Caliper directly to transform Gram-positive bacteria.
