The LabChip GX
software controls the LabChip GX and LabChip GX II instruments. The LabChip GX
software is also used to analyze and compare data in the form of gels or graphs
across different wells from the same or different LabChip GX data files. The
software can also compare data with similar characteristics by applying filters.
Results can be exported to graphic files or .csv (comma separated values) files.
The software can be installed in Reviewer mode to be used only for data
analysis. The 21 CFR Part 11 option can be installed on the LabChip GX computer
to comply with 21 CFR Part 11 requirements. The 21 CFR Part 11 option supports
remote data backup to a database on a network server. Use the CDR Server Utility
software to set up the remote server.

To install the
LabChip GX software on Windows XP:
1.
Close all open programs.
2.
Insert the LabChip GX Installation CD into the CD-ROM drive.
3.
Follow the instructions in the setup wizard.
If
the installation does not automatically start:
1.
From the Start menu, select Run.
2.
Type d:\setup.exe (or type the
drive letter of the CD-ROM drive).
3.
Click the OK button.
4.
Follow the instructions in the InstallShield
wizard.
To
install the LabChip GX software on Windows Vista (Reviewer Mode
ONLY):
1.
Close all open programs.
2.
In Windows Explorer, right-click on setup.exe and select “Run as
administrator.”
3.
In the Warning dialog, select the Allow option. The InstallShield Wizard
displays the list of prerequisites.
4.
Click the Install button to install the required prerequisites. The LabChip GX
InstallShield Wizard opens.
5.
Follow the instructions in the InstallShield wizard.
To install or set
up the remote CDR, you must log onto the Remote Server computer as a Windows
administrator.
For security
purposes, files and folders on the remote server pc should not be shared or
accessible by regular users on the network.
If Subversion V1.5
or higher is installed on the remote server, a dialog prompts you to manually
uninstall subversion. Installation will not continue until Subversion is
uninstalled. After manually uninstalling Subversion, restart the
installation.
To install the CDR
Server Utility software on Windows XP:
1.
In Windows Explorer, double-click on
CDRServerUtilitySetup.exe.
2.
Follow the instructions in the InstallShield
wizard.
3.
If this is the first time the CDR Server Utility is installed, you must restart
the computer.
To
install the CDR Server Utility
software
on Windows Vista:
1.
In Windows Explorer, right-click on CDRServerUtilitySetup.exe and select “Run as
administrator.”
2.
In the Warning dialog, select the Allow option. The CDR Server Utility
InstallShield Wizard opens.
3.
Follow the instructions in the InstallShield wizard.
4.
If this is the first time the CDR Server Utility is installed, you must restart
the computer.
To
start the CDR Server Utility software, right-click on the desktop icon and
select ‘Run as Administrator.’
Troubleshooting
Remote CDR Server installation:
1.
After creating the server, verify the windows service ‘Subversion’ is
running.
2.
The Remote and Local computers should be in the same workgroup. If they are not
in same workgroup, use the IP Address instead of the computer name when
specifying the remote CDR server on the LabChip GX
computer.
3.
After creating the server and folder, if the Get Folder Details check box was
selected, the server name, folder name, username, and password are saved in a
text file in “C:\Program Files\Caliper Life Sciences\CDRServer\ServerDetails”.
This information is required for backing up the local folder on the LabChip GX
computer to the remote CDR folder.
To
uninstall the LabChip GX software:
1.
From the Start menu, select Control Panel.
2.
Double click on Add or Remove
Programs in the Control Panel.
3.
Select LabChip GX from the list of
installed programs.
4.
Click on the Remove button to uninstall the LabChip GX
software.
New
Features:
· Glycan Assay – New
assay for identifying and measuring relative quantities of Glycans on LabChip GX
II instruments. The Glycan ladder is a series of molecules assembled as
multiples of a primitive Glycan molecule = 1 Caliper Glycan Unit
[CGU].
· Default Ladder –
Provides ability to select a ladder as the default ladder that can be applied to
any group of wells in the plate. Default Ladder is exported in assays and can be
used in subsequent runs.
· Column-wise
Sipping – Provides ability to select the order in which the wells are sampled.
Select Column-wise in the Start Run window to sample by column (A1, B1, C1…).
Select Row-wise in the Start Run window to sample by row (A1, A2, A3…). (Ladders
are sipped after every 12 wells, regardless of the sipping
order.)
· Titer Analysis
Improvements – Ability to select either Linear fit or Quadratic Fit for Titer
Curve. Force Fit Through Origin option now available to force Titer Curve
through (0,0) to improve accuracy of calculations for small peaks.
· Filter Legend in
Gel View – Show Filter Legend in Gel Properties provides ability to display a
legend of Filter colors in Gel View.
· Include Peak
Option – Ability to include peaks outside marker (Peak Type is “?”), which are
normally excluded. Included peak will be quantified and included in sample peak
area/concentration totals.
· Improved Column
Selection in Peak Table and Well Table – Right-clicking on the column header in
the Peak Table View or Well Table View opens the Select Peak/Well Table Columns
window opens to choose the columns to display in the
table.
· Raw Data Export
in NetCDF Format – Enables export of Raw Data in NetCDF Format (formerly AIA format). Generates one .cdf file for each selected well.
· Print
Improvements – Add Border option now available for Peak Table or Well Table.
Adds borders between columns and wells in printed tables. Well Header option
available for Peak Table to add well identifier above peak table for each well.
Column Header option available for Peak Tables. Per Page option repeats column
headings at the top of each page. Per Well option repeats column headings at the
beginning of each well. When Peak and Well Tables are printed together, the
tables are combined and printed on one page per
well.

Issues:
Open
Issues in LabChip GX V2.2:
·
Changes to the Default Ladder may
not be logged in Version History. (LCF-3584)
·
Plates sipped with LabChip GX V2.2
software using column-wise sipping order do not display some ladders when viewed
in LabChip GX V2.1 software. (LCF-3577)
·
Default Ladder is identified as
“Ladder 9” in the Version Log. (LCF-3553)
·
All viewers are hidden if the
LabChip GX V2.2 software is minimized during the run. Do not minimize the
software during a run. (LCF-3546)
·
Peaks excluded using the Excluded
Peaks Tab in the Assay Analysis window display the "Include Peak" option on the
right-click menu in the Graph View or Peak Table View. Selecting the “Include
Peak” option does not include the peak. Remove the peak from the Excluded Peaks
table to include the peak in the analysis.

Data File Version Details displays
unnecessary information when updating files from V2.1 to V2.2.Settings for new
options display as changes. (LCF-3673)
·
Electropherogram View displays a
blank window after pressing Ctrl+Z to Undo. Select the
well again in another view to display the electropherogram. (LCF-3624)
·
Front panel buttons may not function
after terminating the software using CalTaskClean.
Restart the instrument or restart the LabChip GX software to reactivate the
front panel buttons on the instrument. (LCF-1828)
·
CalTaskClean closes any open
document or window that starts with "LabChipGX", not
just the LabChip GX software. (LCF-3418)
·
Selected peak in peak table may not
be visible in overlay electropherogram if there are
other peaks near it. (LCF-3273)
·
"USB Key not found" error may occur
while starting the software if the USB cable is disconnected in between a run
and then connected. Workaround: Unplug the cable again, plug it back in, wait a
few seconds, and then restart the software.

New
Features:
· High Size
Filtering – Provides ability to use a shorter filter for smaller size peaks at
the beginning of the well and a longer filter for larger size peaks at the end
of the well. This feature prevents “ringing” of smaller size peaks when using
longer filter for entire well.
· Merge Peaks –
Provides ability to combine two adjacent peaks into a single peak by excluding
one peak and extending the baseline of the remaining peak to include the
excluded peak.
· Smear Analysis –
Provides ability to define a range of data between two specified sizes. Smears
display in the Well Table view. Smears are similar to peaks and have many of the
same properties.
· Titer – Ability
to define a set of wells containing a molecule of known concentration. The range
of concentrations provide a calibration curve from computing sample
concentrations from the peak area. The titer replaces the Ladder well as the
calibration standard for the area to concentration
conversion.
· Auto Export
Contrast Lane – Auto Export settings now include ability to select a specific
gel lane to use to set minimum and maximum contrast for all exported gels.
· Filter AND
Options – Two filter AND options are now available: AND (Well) and AND(Peak).
AND(Peak) replaces the AND option available in previous versions. AND(Peak)
finds any well that contains a single peak that matches all selected filters.
AND(Well) finds any well that contains peaks that match the selected filters.
The peaks that match do not have to be the same
peak.
· Collection Undo
Option – Provides ability to undo changes to Viewer locations, Split locations,
Viewer properties, and Filter list. (Does not undo changes to or affect loaded
plates, Analysis settings, Peak overrides, or Manual well
selections.)
· Print
Enhancements – Print the Electropherogram and peak table/well table of a single
well on the same page. Printing E-Gram + Peak Table or E-Gram + Well Table in
Portrait interleaves the peak Table/Well Table with the Electropherogram. Reduce
the height of the Electropherogram graph in interleaved printing to 44% of the
page height.
· Service Access
Level in CFR Mode – Added Service Access Level in 21 CFR Part 11 mode. By
default, Service access level has permission to Run Assay and Perform
Validation.
· Audit Trail
Search in CFR Mode – Search options added for searching Audit Trail:
1.
Search most recent entries: by
default lists latest 2000 records
2.
Search by dates: lists records within the
selected period
3.
Search entire database: lists all
records from database
4.
Can now search using User Name,
Filename, or Event Category.
· Audit Trail
Manage Columns – Provides ability to select columns to view in Audit Trail list
by selecting check boxes in Manage Columns Window. Also provides ability to set
display order of columns. (Removed multiple views from Audit Trail Window. Now
only provide one view.)
