| Measurement | The 10mM stock solutions (in DMSO) of test samples and/or reference compounds are added (diluted) in universal buffer (pH 7.4) in a 96-well filter plate. The resultant solution and/or suspension in the plate are shaken for 90 minute at ambient temperature. The filter plate is then placed on top of a collection plate, and the assembly is centrifuged with a table top centrifuge (@1,500rpm) for about 3 minutes. The concentration of test sample (or reference compound) in the filtrate is measured spectrophotometrically (UV/Vis) and compared with known concentration of standard controls. Compounds are run at 8 concentrations in duplicate. |
