CDAS EpiScreen is a comprehensive suite of epigenetic targets services designed to support hit discovery and lead optimization efforts. Most of our assays are developed in a Caliper assay LabChip® format and are fully optimized to deliver superior data quality and excellent data reproducibility.
HTS
• Screening conditions tailored to your needs (plate layouts, control wells, etc.)
• High QA & QC standards (postive and negative controls run throughout all the plates)
• Automated compound cherry picking capabilities for hit compound confirmation
• Complete standard or customized report for seamless integration into in-house databases (IDBS…)

IC50 Evaluation - 12 point or custom duplicate IC50 curves
• Choice of dose range of concentrations
• Quality control: reference compound IC50 determination and comparison to historical database
• Low amount of compound required
• Turnaround time: 2 weeks

Mechanism of Action (MoA) Studies
Provides detailed information about the manner in which a drug interacts with enzymatic targets. MoA studies and particularly important for characterizing the specific type of enzyme inhibition and for assessing kinetic on and off rates.

LabChip Technology platform
CDAS conducts most of its EpiScreen services using the LabChip microfluidic technology developed by Caliper Life Sciences.
Key features and benefits of the technology:
• No monoclonal antibodies or radioisotopes are required
• Detection of biochemical inhibition in a direct, homogenous, ratiometric, high quality format
- substrate and product are measured in every well
The separation and direct measurement of substrates and products allowing:
• Higher signal-to-noise ratios
• Fewer false positive and negative results
• Identification & elimination of enzymatic activities not associated with the kinase reaction of interest
• The ability to read over time (kinetic format)
A small reaction volume is used for the detection of substrate and product:
• Minimum sample requirements (100 ug) is enough for profiling a compound in a panel of assays
• Profile your new lead compounds and other chemicals faster at a lower cost
• Due to minimal sample requirements you save your compound for additional CDAS in vitro studies
