Three (3) assays performed over a four-week time period are used to assess the sexual virility and fertility of male mice. The block includes the evaluation of fertility, erectile function and sexual performance of genetically modified or pharmacologically treated mice. For fertility measures, the male mice are placed with gonadally intact females for one week; the presence of a vaginal plug, latency to the plug, pregnancy, and litter size are recorded for each mouse. To ensure the best possible sexual performance, each male undergoes additional sexual training prior to the contact sexual behavior assay. During the two-week training period, the mice are allowed four exposures to primed ovariectomized females to further increase their sexual experience. After the training period, the mice are tested for mechanically-induced erectile function and then their sexual virility is measured in the male contact sexual behavior assay. Each mouse is placed with a primed ovariectomized female and observed for 45 minutes. The number of mounts, intromissions, and ejaculations and the latency to the first of each of these behaviors is determined.
