Multi-label imaging without the cross-talk


Today’s biological imaging demands in-depth study of extremely complex systems, and often requires multi-label analysis. Caliper’s multispectral microscopy imaging systems enable quantitative evaluation of multiple fluorescent and brightfield molecular markers simultaneously even when co-localized in a single tissue section. The data for each marker is spectrally unmixed, generating clear and accurate images for each of the individual labels in a multi-labeled tissue section with no cross-talk.  

Our microscopy imaging systems deliver the high quality data provided by flow-cytometry, but unlike this technique, the morphological context is preserved down to the sub-cellular level. In addition, autofluorescence is completely removed, increasing signal-to-noise by up to 100x.  Low signal levels that were previously undetected can now be captured, and overall quantitation is vastly improved - particularly for solid, formalin-fixed, and paraffin-embedded tissues.

Caliper’s microscopy imaging systems are ideal for the following areas of research:

  • Microenvironment analysis, including assessment of tumor immunogenicity
  • Tissue cytometry, including multiplexed per-cell measures of nuclear, cytoplasmic, and membranous protein expression
 

Brightfield immunohistochemistry with color re-mapping for legibility. Oral squamous carcinoma (FFPE, 20X) stained for CD34 (endothelium, Nova Red) and podoplanin (VIP, purple), and counterstained with hematoxylin. The spectral dataset was unmixed, the component planes re-colored (purple to green) and displayed as a composite image with improved contrast and legibility over the original.

Caliper’s microscopy imaging systems are an easy add-on to any microscope with a camera port, and enable simple image acquisition and high-contrast separation of multiple chromogens or fluorophores.  Both high-resolution color and multispectral images are captured, and can be exported in standard image file formats. In addition, Nuance systems are compatible with Molecular Devices' MetaMorph™ software.
