NCI-N87-luc2-GFP Bioware® Ultra Green
NCI-N87-luc2-GFP is a luciferase and green fluorescent protein (GFP) expressing cell line. This cell line can be used to establish:
- Orthotopic tumor models of gastric carcinoma
- Subcutaneous tumor models
- Ex vivo cell isolation using fluorescent activated cell sorter (FACS)
- Post genomic analyses of isolated tumor cells.
- Immunohistochemistry detection of GFP positive cells

In vitro Luciferase and GFP stability

500,000 cells were plated per well in black-walled 24-well plates. Cells were incubated for 37C for recovery overnight and luciferase assay was performed using an IVIS Spectrum. Each experiment was done in quadruplicates. The assays were repeated over four weeks period of time. Bioluminescence data was analyzed using the Living Image 4.0 software.
Growth curve of NCI-N87-luc2-GFP

1 x105 cells were plated on T25 flasks and the total numbers of cells were counted every 24 h using a Nexcelom automatic cell counter.
Subcutaneous Tumor Growth in a Nu/nu Mice

1 x 106 NCI-N87-luc2-GFP cells were injected subcutaneously into the dorsal region near the thigh of female nu/nu mouse. Tumor growth was monitored by IVIS Spectrum at various time points for both luciferase and GFP expression. For luciferase expression, mice were imaged 10 minutes post i.p. injection of Luciferin at 150mg/kg at various time points. The image above shows tumor growth from representative mouse.
