pXen05 (pAUL-A Tn4001 luxABCDE KmR)
This is a transposon-based lux delivery vector that contains a Gram-positive optimized luxABCDE cassette. The inverted repeats (IR) of the Tn4001 transposon are located at positions 305-332 and 8161-8188. The luciferase genes luxA (374-1453), luxB (1475-2455), luxC (2478-3917), luxD (3940-4860), luxE (4883-5992), and kanR (6016-6807) are co-transcribed.

This plasmid is a shuttle vector that will replicate in a range of Gram-positive and Gram-negative bacteria. However, amplification of this plasmid in E. coli will induce mutations and make the plasmid unstable. Thus, we recommend using the plasmid supplied from Caliper directly to transform Gram-positive bacteria.
